2 . Mitochondrial function in leptin - deficient obese - hyperglycaemic mice [ ob / ob mice ]

چکیده
This review summarises key aspects of what has been learned about the effect of leptin on mitochondrial function from studies in leptin deficient obese ob/ob mice. Ob/ob mice are grossly overweight and hyperphagic particularly at young ages. The feeding behaviour in combination with leptin deficiency leads to severe insulin resistance with hyperglycaemia and hyperinsulinemia. Mitochondrial metabolism is important in the regulation of insulin release from pancreatic ß-cells. Ob/ob mouse ß-cells respond adequately to most stimuli and ob/ob mice have been used as a rich source of pancreatic islets with high insulin release capacity. Mitochondrial metabolism is altered in leptin deficient animals leading to reduced oxidative capacity in liver and muscle cells. Uncoupling proteins are down regulated in leptin deficient mice and the production of reactive oxygen species is increased.

Impaired cardiac contractile function in ventricular myocytes from leptin-deficient ob/ob obese mice.
The level of the obese gene product leptin is often positively correlated with body weight, supporting the notion that hyperleptinemia contributes to obesity-associated cardiac dysfunction. However, a link between leptin levels and cardiac function has not been elucidated. This study was designed to examine the role of leptin deficiency (resulting from a point mutation of the leptin gene) in ca...
متن کاملAltered Hepatic Lipid Metabolism Contributes to Nonalcoholic Fatty Liver Disease in Leptin-Deficient Ob/Ob Mice
Nonalcoholic fatty liver disease (NAFLD) is strongly linked to obesity, insulin resistance, and abnormal hepatic lipid metabolism; however, the precise regulation of these processes remains poorly understood. Here we examined genes and proteins involved in hepatic oxidation and lipogenesis in 14-week-old leptin-deficient Ob/Ob mice, a commonly studied model of obesity and hepatic steatosis. Obe...
متن کاملAdrenalectomy reverses obese phenotype and restores hypothalamic melanocortin tone in leptin-deficient ob/ob mice.
In genetically obese leptin-deficient ob/ob mice, adrenalectomy reverses or attenuates the obese phenotype. Relative to lean controls, ob/ob mice also exhibit decreased hypothalamic proopiomelanocortin (POMC) mRNA and increased hypothalamic agouti-related peptide (AGRP) mRNA and neuropeptide Y (NPY) mRNA. It has been hypothesized that this profile of hypothalamic gene expression contributes to ...
متن کاملIdentification of major quantitative trait loci controlling body weight variation in ob/ob mice.
The adipocyte hormone leptin constitutes an important component of the regulation of energy homeostasis; leptin-deficient animals, such as obese mice, are strikingly overweight. The seemingly uninhibited weight gain in obese mice belies the fact that control of energy homeostasis remains under precise, heritably modifiable control. Herein, we report large, heritable differences in body weight a...
متن کاملObese and diabetic db/db mice develop marked liver fibrosis in a model of nonalcoholic steatohepatitis: role of short-form leptin receptors and osteopontin.
Obesity and type 2 diabetes are associated with nonalcoholic steatohepatitis (NASH), but an obese/diabetic animal model that mimics human NASH remains undefined. We examined the induction of steatohepatitis and liver fibrosis in obese and type 2 diabetic db/db mice in a nutritional model of NASH and determined the relationship of the expressions of osteopontin (OPN) and leptin receptors to the ...
